Stem Cell Transplantation in Umbilical Cord Blood(I) Expansion Effects of Stem Cells in Umbilical Cord Blood with Various Hematopoietic Growth Factors

Abstract
Stem cell transplantation is used for the patients lacking the capability of production of hem-atopoitic cells. Recently, there have been a few studies using the umbilical cord blood as a sou-rce of stem cells, however it remains unclear whether umhhilical cord blood contains sufficient amount to reconstitute the patient. Our study was designed to know that umbilical cord blood is proper material for stem cell tr-ansplantation. We measured colony formation and CD34(+) cell in umbilical cord blood with ad-dition of hematopoietic growth factors(G-CSF, GM-CSF, SCF, IL-3, and GM-CSF) +SCF +IL-3). The results were as follows: 1) There are significant correlation between colony froming cells and the cells expressing C D34(+) which are immunophenotype of stem cells(r=0.88, p=0.02). 2) Hematopoietic stem cell in umbilical cord blood were differentiated into CFU-GM, CFU-G, and CFU-M and expanded with addition of the hematopoietic growth factors, especially with co-mbination of GM-CSF, SCF, and IL-3(p<0.05). 3) After incubation with GM-CSF, CD34(+) cells were expanded which counted 3.62 times o n 3rd day and 11.84 times on 7 day(p<0.05). In this study, we found that stem cells in umbilical cord blood may differentiated into various forms of hematopoietic cells and the expansion is promoted by growth factors. This observation raised the possibility that umbilical cord blood is a candidate of stem cell transplantation.
