Abstract
Eberlein's simple partition method is in general use for the measurement of free bilirubin, bilirubin mono and diglucuronide which is important in the evaluation of neonatal jaundice. In this method human serum albumin is used for the preparation of standard bilirubin solution, but the human albumin is expensive and not available commercially in this country. Therefor, authors substituted human albumin with unhemolysed pooled human serum from healthy donors and compared the results with Eberlein’s method. In the author’s method the maximum absorption spectrum of UE and UB fractions is identical to that of Eberlein's method but the spectrum of LB is 430 m/z. The bilirubin concentration and the optical density show straight line correlation concordant with the Lambert-Beer’s law. The bilirubin value by the author’s method corresponds well with that of Eberlein’s method. The maternal bloods . revealed free bilirubin, bilirubin mono- and diglucuronide values of 0.564±0.266, 0,256±0.044 and 0.924±0.184; the cord bloods 1. lll±0.110, 0.362±0.049 and 0.832±0.053 mg/100 ml respectively by the author’s method.
