Streptozotocin produces diabetes in laboratory animals through the destuction of the pancreatic beta cells. The diabetogenic activity of STZ, however, is known to be modified by several agents such as nicotinamide, diazoxide, adrenergic blockers and petussis vaccine (PV). The present study was undertaken to observe the protective effect of PV or boiled pertussis vaccine (bPV; incubating the PV at 80 C for 30 mintues) against the development of insulin dependent diabetes induced by STZ in young rats, and also the effect of PV on the activites of glucokinase & glucose 6-phosphatase in the liver. PV at a dose of 1.2xIO10 miroorganism was administrated on 3days before & 7 days after a single injection of STZ (60 mg/kg body wt). The blood glucose and insulin levels were measured at 1st, 2nd, 4wks, 6th and 8th wks after STZ injection, and the activities of glucokinase & glucose 6-phosphatase and the histological examination were studied at 8th wk after STZ 1 injection. The following results were obtained. 1) No significant difference was noted in blood glucose levels between STZ group and STZ+PV or STZ+bPV groups (p>0.05). 2) Blood insulin levels in STZ group decreased significantly from 2nd wk (p < 0.05). Compared with STZ group, blood insulin levels in STZ + PV & STZ+bPV groups were higher, not a significant value except at 2nd wk in STZ + PV group(p>0.05). 3) No significant difference was noted in the activities of hepatic glucokinase & glucose-6 phos- phatase between STZ group and STZ + PV or STZ+bPV group(p>0.05). 4) On microscopically, a destruction of Langerhan’s islet and its cells in STZ + PV and STZ+bPV groups was less remarkable than that in STZ group. These results suggest that the PV has somewhat protective effects on islet cell of pancreas in rats but it could not reduced blood glucose levels significantly.
